If you are at Cancun airport, the best option to make the trip by land is through a private transfer service to Isla Mujeres, which as such has the objective of taking you and your companions from Cancun airport to the Puerto Juarez maritime terminal.
The maritime terminal of Puerto Juarez is the busiest terminal in the city and the preferred one for all the people who want to visit Isla Mujeres, because every 30 minutes ferries depart to Isla Mujeres.
The first transfer service to Isla Mujeres is at 6:00 AM and the last check out time is at 11:30 PM and the approximate duration of the transfer is about 30 minutes.
Once you arrive at Isla Mujeres, you can take a cab that costs approximately 50 to 100 Mexican pesos (2.5 USD to 5 USD) to get to your hotel.
